STUDI PERBANDINGAN FOSFOLIPID MEMBRAN
DARI ERITROSIT NORMAL DAN PENDERITA
HIPERLIPIDEMIA

RINGKASAN
Peningkatan metabolisme lipid akan menimbulkan pe-
ningkatan kadar lipid darah atau hiperlipidemia yang se-ring
dihubungkan dengan penyakit jantung koroner, aterosklerosis,
zamtomatosis, dan pankreatitis. Kolesterol, ester kolesterol,
trigliserida dan fosfolipid merupakan lipid utama yang terdapat
di dalam darah. Analisis hubungan antara hiperlipidemia,
khususnya kolesterol dan trigliserida dengan jenis-jenis
penyakit di atas telah banyak dilakukan di dalam bidang
kedokteran, sedangkan studi tentang pengaruh fosfolipid belum
banyak dilakukan, terlebih-lebih pengaruhnya terhadap kandungan
fosfolipid membran eritrosit itu sendiri. Dalam studi
penelitian ini peneliti mencari korelasi antara komponen-
komponen fosfolipid dalam membran eritrosit normal dan membran
eritrosit penderita hiperlipidemia, baik secara kualitatip
maupun secara kuantitatip. Untuk mencapai hal itu telah
dilakukan analisis kasus terhadap 30 sampel darah penderita
hiperlipidemia dan 30 sampel darah normal pada berbagai usia.
Ekstraksi lipid total dari membrane eritrosit di-
kerjakan berdasarkan metode Steck dan Kant, analisis kualitatip
dilakukan secara kromatografi lapis tipis sistem satu dimensi,
sedangkan untuk analisis kuantitatip
digunakan gabungan metode kromatografi lapis tipis de-
ngan spektrofotometri. Dari hasil penelitian, secara kus-
litatip, telah ditunjukkan bahwa macam komponen fosfolipid
membran eritrosit penderita hiperlipidemia tidak berbeda dari
pada fosfolipid membran eritrosit normal, sedangkan dari hasil
analisis kuantitatip dengan pengolahan data statistik diperoleh
peningkatan kadar fosfatidilkolin dan fosfatidiletanolamin
sedangkan untuk sfingomielin diperoleh sebaliknya. Perbed.aan
ini cukup memberikan arti yang bermakna.
ABSTRACT
Hyperlipidemiae, elevation in blood lipids, may occur
by alteration in lipid -metabolism, frequently correlated
with coronary heart discase, atherosclerosis, zamtomatosis
or pancreatitis. The major lipids in the blood are
cholesterols, ester cholesterols, triglycerides and
phospholipids.
The relationship of hyperlipidemiae, particularly
cholesterol and triglycerides, to those disease had been
observed, but a study of the influence of the phos-
pholipids, particularly upon the membrane of the erythro-
cyte, have not well described yet.
The research was done to investigate the correlation
between the phospholipids of the normal erythrocyte mem-
brane and the hyperlipidemic one quantitatively and quali-
tatively.
Thirty blood samples from hyperlipidemic patients and
thirty blood samples for normal patients of different ages
were used in this research.
The extraction of total lipids from the membrane ery-
throcyte was carried out according to Steck and Kant
method.
One dimensional thin layer chromatography was used in
the qualitative analysis, and the combination of thin
layer chromatography and spectrophotometry was used in
the quantitative analysis.
The results showed that the number of individual
phospholipid components from those two erythrocyte mem-
brane were the same. However, quantitative analysis of
those phospholipids showed that both phosphotidyl choline
and phosphotidyl ethanolamin were significantly different.
