• Serabut preganglionik Acethylcholine• Serabut postganglionik :
– Simpatis Norepinephrine (Levarterenol)
– Parasimpatis Acethylcholine
• Organ yang diinervasi umumnya memiliki mekanisme regulasi intrinsik
Denervasi penurunan fungsi ( kehilangan fungsi)
• Memiliki lengkung refleks
• Berfungsi regulasi organ - organ di luar kontrol kesadaran

Neurotransmisi
• Konduksi aksonal Hantaran impuls listrik sepanjang akson
• Membran aksonal pada Resting membrane potential :– Relatif permeabel terhadap K+ (Kation intrasel
terbanyak)
– Impermeabel terhadap Na+ & Cl- (ion ekstrasel terbanyak)
• Resting membrane potential dipertahankan oleh aktivitas Na+/K+ ATP-ase yang diaktivasi oleh pe Na+ intrasel / pe K+ ekstrasel
Axonal Conduction
• Phase I : – Downstream impulse threshold level perubahan
konformasi kanal ion permeabilitas Na+ me transient rapid influx Na+ positive overshoot Voltage - sensitive Na+ channels
• Phase II :– Delayed activation K+ channel efflux K+
hyperpolarization aktivasi Na+/K+ pump (Na+/K+ ATP-ase) resting state
• Selama potensial aksi, depolarisasi dapat memanjang dengan aktivasi Ca2+ channel slow inward Ca2+ current
• Perubahan polaritas membran akson mempengaruhi kanal ion pada akson di dekatnya propagasi impuls sepanjang akson akson terminal
Junctional Transmission
• Impuls akson terminal, menyebabkan :– Pelepasan transmitter dalam vesikel dari akson terminal
eksositosis
– Ikatan transmitter – postjunctional receptors perubahan postjunctional potential membrane aktivasi ligand - gated channel
• EPSP (Exitatory Postsynaptic Potential) (Nicotinic receptors, Glutamate, 5-HT, purine receptors) peningkatan konduktansi Na+
• IPSP (Inhibitory Postsynaptic Potential) (GABA, glycine receptors) peningkatan konduktansi Cl-
– Perubahan aktivitas postjunctional cell
– Destruksi / pengalihan transmitter dari junctional sites
Transmisi Kolinergik• Sintesis Asetilkolin
– Choline Acetyltransferase enzim yang mengkatalis reaksi akhir sintesis ACh.
– Choline + Acetyl-CoA ACh. (berlangsung di sitoplasma)
• Transpor Choline• High-affinity transport system
• Low-affinity transport system
– High - affinity system khas pada neuron kolinergik
– Uptake choline ke dalam axoplasma dihambat oleh Hemicholinium
– ACh dikemas dalam vesikel ditranspor ke ujung saraf dihambat oleh Vesamicol
Pelepasan & Terminasi ACh• Potensial aksi influx Ca2+ terikat dg vesikel
eksositosis
• Pelepasan ACh berlangsung dalam jumlah konstan (kuanta)
• Acetylcholine esterase (AChE) hidrolisis ACh dari synaptic gap
Tipe Reseptor & Transduksi Sinyal• Reseptor Nikotinik ligand - gated ion channel
aktivasi selalu menyebabkan peningkatan permeabilitas Na+ & Ca2+
• Reseptor nikotinik dikelompokkan dalam 2 subtipe :
• subtipe (8 subtipe, 2 – 9) & (3 subtipe, 2 – 4)
• Reseptor Muskarinik merupakan G protein - coupled receptor
• M1, M3, M5 aktivasi protein G (Gq/11) stimulasi aktivitas phospholipase C
• M2 & M4 berinteraksi dengan protein G yang berbeda, (Gi & G0) hambatan adenilat siklase, aktivasi K+ channels, supresi aktivitas voltage - gated Ca2+ channels

• MAO & COMT, tersebar luas pada berbagai bagian tubuh, termasuk CNS; Konsentrasi terbesar di liver & ginjal
Reseptor Adrenergik• Reseptor adrenergik dikelompokkan ke dalam :

• Potensi agonis katekolamin endogen terhadap tipe reseptor:
